چکیده

Milstein, Adam B. Ph.D., Purdue University, August, 2004. Imaging of near-infraredfluorescence, absorption, and scatter in turbid media. Major Professor: Charles A.Bouman and Kevin J. Webb.A nonlinear, Bayesian optimization scheme is presented for reconstructing fluo-rescent yield and lifetime, the absorption coefficient, and the diffusion coefficient inturbid media, such as biological tissue. The method utilizes measurements at boththe excitation and emission wavelengths for reconstructing all unknown parameters. The effectiveness of the reconstruction algorithm is demonstrated by simulation andby application to experimental data from a tissue phantom containing the fluorescentagent indocyanine green.

Near-infrared narrow-band imaging of gold/silica nanoshells in tumors.

Gold nanoshells (GNS) are a new class of nanoparticles that can be optically tuned to scatter or absorb light from the near-ultraviolet to near-infrared (NIR) region by varying the core (dielectric silica)/shell (gold) ratio. In addition to spectral tunability, GNS are inert and bioconjugatable, making them potential labels for in vivo imaging and therapy of tumors. We report the use of GNS as ...

Imaging through scattering media by the use of an analytical model of perturbation amplitudes in the time domain.

A method of generating images through highly scattering media is presented that involves comparing measurements of the time-dependent intensity of transmitted light with an analytical model describing the sensitivity of that intensity on localized changes in optical properties. A least-squares fitting procedure is employed to derive the amplitudes of the measurement perturbations caused by embe...

Plasmonic Nanostructures and Applications

Gold nanoparticles with unique optical properties may be useful as biosensors in living whole cells. Using a simple and inexpensive technique, we recorded surface plasmon resonance (SPR) scattering images and SPR absorption spectra from both colloidal gold nanoparticles and from gold nanoparticles conjugated to monoclonal anti-epidermal growth factor receptor (anti-EGFR) antibodies after incuba...

The effects of internal refractive index variation in near-infrared optical tomography: a finite element modelling approach.

Near-infrared (NIR) tomography is a technique used to measure light propagation through tissue and generate images of internal optical property distributions from boundary measurements. Most popular applications have concentrated on female breast imaging, neonatal and adult head imaging, as well as muscle and small animal studies. In most instances a highly scattering medium with a homogeneous ...

Optical images from Pathophysiological signals within breast tissue using three-dimensional near-infrared light

Near Infrared (NIR) tomography has the potential for characterization of different tissue types based upon cellular and vascular alternations that are optically apparent. This is especially useful for characterizing cancerous regions within normal tissue due to the high available contrast. Reconstructed images from NIR light propagation measurements through the female breast hold promise of pro...
